Product Description

This product is a powdered McCoy's 5A medium containing 1.5 mM L-alanyl-L-glutamine with phenol red indicator, no HEPES, no sodium pyruvate , and additional sodium bicarbonate for use.

Storage and Shipping Conditions

Ship at room temperature; Store away from light at 2-8℃, valid for 3 years.

General guidelines

1. Prepare clean container close to final volume.

2. Prepare distilled water at room temperature (15-30°C) or water for cell injection (recommended G4701).

3. Prepare 1 N NaOH and 1 N HCl.

Procotols of liquid media preparation from powder

1. Fill the prepared clean container with 95% final volume of distilled water, take 1 bottle of this product, or weigh the powder according to the proportion and put it into the water to dissolve. Note that the powder can be dissolved quickly after disintegration in water, if high-speed stirring may cause the powder to form a ball, the surface of the formation of a water film, but not conducive to dissolution. Therefore, it is not necessary to stir the powder at high speed after putting it into water..

2. After the powder is dissolved and clarified, add sodium bicarbonate at 2.2 g/L and stir to dissolve. If other volumes are required, add the corresponding amount of sodium bicarbonate according to the corresponding volume.

3. Adjust the pH with 1 N NaOH and 1 N HCl to 0.2-0.3 units below the operating pH (recommended operating pH 7.0-7.4). The solution will increase in pH by 0.1-0.3 units after filtration.

4. Add water to final volume. Keep the solution sealed before filtration.

5. Positive pressure filtration using a 0.2 μm membrane into a sterile container for backup (note aseptic handling).

6. At the end of filtration, a little liquid can be taken for aseptic examination, and used after qualified. Prepared liquid culture medium is stored at 2-8℃ with a shelf life of 1 year.
